Jill Pecon-Slattery, PhD, is the Founder and CEO of Cornerstone Genomics and the scientific force behind CodeXome®—a platform that unites evolutionary genomics, AI, and protein biology to transform how researchers interpret variants, discover targets, and understand disease. A comparative genomicist by training, Jill spent more than two decades at the National Institutes of Health studying how genes evolve, how proteins tolerate change, and how natural selection shapes the limits of biological function. Her work is grounded in a simple but powerful belief: biology tells its story across millions of years, and if we listen, we discover answers modern tools have missed. To enable this, Jill led the creation of the world’s most comprehensive primate exome resource—19,244 genes sequenced across 55 non-human primate genera, producing ~2 billion datapoints of evolutionary history that reveal how each gene changes, which amino acid positions are functionally constrained, and where natural variation is safely tolerated. This “Deep Time Ancestry” database forms the backbone of CodeXome, allowing users to: • filter out natural, benign variants in a single step,
• reclassify VUS with evolutionary evidence,
• interpret functional motifs and structural domains,
• identify unique human-specific changes linked to disease,
• and predict protein impact using evolutionary AI/ML models. Her validation studies with ClinGen and the BRCA Exchange showed something foundational: pathogenic variants do not co-occur with natural variation across primates, while benign variants consistently do. These findings confirmed the central premise of CodeXome—evolution has already run the perfect natural experiment.
